| I. Background | |
| PACAP/VIP 受體家族屬于 B 類 G 蛋白偶聯(lián)受體(GPCR),包含 PAC1R (PAC1)、 VPAC1R (VPAC1) 和 VPAC2R (VPAC2) 三個亞型,共同響應(yīng)神經(jīng)肽 PACAP(垂體腺苷酸 環(huán)化酶激活肽)和 VIP(血管活性腸肽)的調(diào)控,通過激活 Gs/cAMP-PKA 核心信號軸 (部分亞型偶聯(lián) Gq/PLC)廣泛協(xié)調(diào)神經(jīng)內(nèi)分泌、代謝穩(wěn)態(tài)與免疫調(diào)節(jié)。 PAC1R(垂體腺苷酸環(huán)化酶激活肽受體 1 型)作為 B 類 GPCR 的核心成員,特異性響 應(yīng)其內(nèi)源性配體 PACAP(垂體腺苷酸環(huán)化酶激活肽)的調(diào)控,通過偶聯(lián) Gs/Gq 蛋白激活 cAMP-PKA 及 PLC-PKC 雙信號軸,主導(dǎo)神經(jīng)可塑性調(diào)節(jié)、垂體激素分泌和疼痛信號傳導(dǎo) 等關(guān)鍵生理過程。PAC1R 在中樞神經(jīng)系統(tǒng)(尤其下丘腦、杏仁核與三叉神經(jīng)節(jié))、垂體前 葉及腸道神經(jīng)元中高表達(dá),其信號異常被證實是多重疾病的核心驅(qū)動機(jī)制:在偏頭痛中介 導(dǎo) PACAP 釋放觸發(fā) CGRP 依賴性神經(jīng)源性炎癥與血管擴(kuò)張,在創(chuàng)傷后應(yīng)激障礙(PTSD) 中通過增強(qiáng)杏仁核突觸可塑性固化恐懼記憶,在耐藥性垂體瘤中則繞過生長抑素受體信號 促進(jìn)腫瘤增殖。 |
